Fibronectin
is a well-characterized, multifunctional cell interactive
protein present in a variety of extracellular matrices.
In a few studies over the last decade, fibronectin
has been reported to influence a variety of melanocyte
biology, including morphology, attachment, proliferation,
migration and pigmentation of melanocyte. It has
been documented that UVA and UVB can induce fibronectin
biosynthesis in hairless mice and normal human skin.
This raises the question about the relationship
between PUVA and fibronectin biosynthesis. This
study was done to investigate the effects of PUVA
on amount of fibronectin and the difference in amount
of fibronectin in supernatants of cultured human
keratinocyte, melanocyte and fibroblast. We measured
the amount of fibronectin by fibronectin enzyme
immunoassay kit in control group and experimental
groups(UVA groups, 8-MOP group and PUVA groups).
The irradiated doses of UVA were 30 mJ/cm2 or 60
mJ/cm2. 8-MOP was added at dose of 2 10-7 M in medium
for 30 minutes. The cells were exposed to 30 or
60 mJ/cm2 of UVA after adding 8-MOP at 2 10-7 M
in medium for 30 minutes in PUVA groups. The results
were as follows : 1. The amount of fibronectin in
supernatants of cultured keratinocyte, melanocyte,
and fibroblast The amount of fibronectin in supernatants
of cultured fibroblast was much higher than the
other cells (keratinocyte, melanocyte)(p 0.01).
The amount of fibronectin in supernatants of cultured
keratinocyte was much higher than melanocyte (p
0.01). The amount of fibronectin in supernatants
of cultured melanocyte was very low. 2. Fibronectin
amount in control and experimental groups There
was no significant difference in fibronectin amount
between control and experiment groups (p 0.05).
In summary, PUVA had no effect on amount of fibronectin
in human skin cells in vitro and the largest amount
of fibronectin was measured in supernatants of cultured
fibroblast. |
